stability
The stability of Nisin is also related to the PH value of the solution. If it is dissolved in skimmed milk with PH=6.5, the activity will only lose 15% after pasteurization at 85 ~C for 15 minutes. When it is dissolved in dilute hydrochloric acid with PH=3, the activity will remain 100% after high pressure sterilization at 121 ~C for 15 minutes. Its acid resistance and heat resistance are excellent. [3]
Bacteriostatic activity
Nisin can effectively inhibit many Gram-positive bacteria causing food spoilage, such as Lactobacillus, Leuconostoc, Micrococcus, Staphylococcus, Listeria, etc. Especially, Nisin has a strong inhibitory effect on spore-producing bacteria such as Bacillus and Clostridium. Generally, spore producing bacteria have strong heat resistance, such as instant sterilization of fresh milk at 135 ℃ for 2 seconds, the mortality of non spore bacteria is 100%, the mortality of spore bacteria is 90%, and 10% of spore bacteria can not be killed. Adding 0.03-0.05g/kg Nisin to fresh milk could inhibit spore germination and reproduction of Bacillus and Clostridium.
Safety
Pathologist studies and toxicological tests have proved that Nisin is completely non-toxic. Nisin can be degraded into amino acids by digestive tract protease, without residue, affecting human probiotics, producing no resistance and cross-resistance with other antibiotics. Many countries in the world, such as Britain, France, Australia and so on, add Nisin to packaged food. Through this method, sterilization temperature can be reduced, sterilization time can be shortened, heat processing temperature can be lowered, loss of nutrients can be reduced, quality of food can be improved, energy can be saved, and storage time of food can be effectively prolonged. It can also replace or partly replace chemical preservatives and chromogenic agents (such as nitrite) to meet the needs of producing health food and green food.
